Once we start editing our genes, where do we stop?

'Genomics' could cure all sorts of diseases - but could also create designer babies. We must debate the implications while we still have time

By Michael Hanlon
7:02PM BST 02 Sep 2015

What is the essence of being a human? Is it our thoughts, our emotions? Are we defined by what we do, our achievements; our great successes and failures? Or are we merely the result of the interactions of some 25,000 genes, segments of DNA that instruct cells to make proteins, contained in what is collectively known as the human genome?

We are all these things, but it is undeniable that at the core of our being lies the genome – the “master code” of any species. Many scientists say that technology now allows us to make improvements – to “edit” this master code and improve upon nature, possibly eliminating genetic diseases such as cystic fibrosis and even improving cognitive function and life expectancy.

It is a seductive idea. This week, however, a call for caution by some of the world’s leading research funders should be heeded. This is not a fatuous argument against “playing God”. Humans have been undoing the work of the Almighty ever since we invented proper medicine and smashed down infant mortality rates.

Nor is it an argument against genetic tinkering per se; there is nothing mystical or sacrosanct about DNA, and the calls to ban the genetic modification of, say, wheat plants to enable them to thrive without the use of harmful pesticides are specious and motivated by silly superstition.

But the human genome is different. It is, quite literally, who we are. Alter the genetic code present in germline cells (sperm and human eggs) and the changes are carried forth into future generations. If this can be used to eliminate diseases such as cystic fibrosis or haemophilia, many say, then surely what is the harm?

Writing in Nature this week, five biomedical funders, including the Wellcome Trust and the Academy of Medical Sciences, call for an “urgent national debate” on the ethics of modifying human embryos and the germline to prevent serious diseases. This may seem moot; after all, the first draft of the human genome was published 15 years ago, and to date the promised “genomic revolution” in medicine has failed to emerge.

But in recent years there has been significant progress in the field of “genomic editing”. A technique called Crispr-Cas9, invented in 2013, uses an enzyme to alter the DNA of cells. This could be used to treat a particular disease in an individual – cancer, say. Most scientists see little danger in gene therapies such as this, as the effects are limited to the recipient.

Using the same technique, however, it is also possible – in theory – to alter, significantly, the genes of germline cells and of embryos. These are the changes that would be passed on. There are two potential problems here: one practical, one philosophical.

The first concerns unintended consequences. Fixing “faulty” genes may, like all treatments, carry side effects. Conventional gene therapy is not without risk; but gene therapy whose risks are made apparent two, or three generations down the line would be a whole different ball game. A patient can consent to treatment; her great-great grandchildren cannot.

Bentley rebuilt from parts scattered around man's home could sell for £800,000

Experts have lovingly restored a classic 1920s Bentley found scattered in pieces throughout the home of its former owner

By Gregory Walton
6:40PM BST 03 Sep 2015

A 1928 Bentley reassembled after its dismantled parts were found scattered throughout a three-storey house and gardens is expected to sell for as much as £1 million.

The parts of the 4.5 litre, art-deco styled vehicle were concealed under beds, in oil-filled glass jars and under a tarpaulin in the owner’s west London home.

The car, just one of eight ever made, was acquired by Stuart Wallace, an English teacher, when he was a student in 1962.

His daughter Bea Wallace-Hartstone called in experts to see if the car could be reassembled following his death.

William Medcalf, head of the Medcalf collection, toured the property scouring for parts until eventually he and his 12 experts had found nearly every original component.

Medcalf subsequently struck a deal with Mrs Wallace-Hartsone, agreeing to buy the parts from the family.

While similar reassembled specimens have sold for £800,000, the firm is tight-lipped about how much they paid for the parts or what they expect to make at auction.

A source said: “You can’t put a price on passion, someone will pay whatever it is worth to them.”

Not one new nut or bolt has been used, with just a handful of non-original wooden supports added to strengthen the chassis.

Mr Medcalf told Octane magazine: “About a year ago a very nice lady called and said her father had passed away and that he’d had a couple of old cars in his house, one of which was a Bentley.”

“She was selling the house and the cars had to go. She explained it was a vintage Bentley but was not absolutely sure it was complete because it was in pieces spread throughout the house.

“So I jumped in a car and drove straight to Kew Gardens. Sure enough, bare on the entrance hall floorboards was a Bentley cylinder block. Going in further I saw a clutch on the stairs, then the connecting rods - there were literally bits everywhere and throughout the three storey house.”

After buying the vehicle for £280, Mr Wallace was deterred by the prohibitively high running costs and decided to store the car in his garage, according to his family.

He went on to strip in and began to restore it but later scattered the component parts throughout his house and garden.

When the Medcalf team began to scour for parts they found pieces deposited in jam jars of oil, the dashboard was concealed in a spare bedroom while the original headlights were found under a bed.

The engine’s original Bentley lead seal was even found to be still in tact.

Mr Medcalf added: “There were pieces everywhere and in every conceivable container.

“We went back a couple of times over the year it took to settle the probate and actually have the components released to us.”

The car’s original aluminum bodywork was later found on the roof of Mr Wallace’s nearby lock-up.

Mr Medcalf added: “As for its value now, that’s when the real debate starts - given its rarity, provenance and originality with matching numbers.

“A four-seater model went for £800,000 a while ago and Bonhams has an original Bentley in its sale at Beaulieu this coming weekend.

Mrs Wallace-Hartstone said: “My dad was always messing around with cars and storing old parts and spares everywhere.

“He kept a dismantled AC Cobra in my bedroom. He was crazy about cars.”

Six Men Charged With Disrupting Flight From San Diego To Chicago

CBS Chicago
September 1, 2015

CHICAGO (CBS) — A Southwest Airlines flight from San Diego to Chicago was diverted to Amarillo, Texas, late Monday night, after six passengers became unruly during the flight.

Southwest flight 1522 was headed to Midway International Airport when a group of passengers traveling together “became disorderly and, subsequently, refused to obey instructions from the crew,” according to a statement from the airline.

An Amarillo Police spokesman said the passengers were fighting and arguing with the flight crew. The airline said the disturbance caused the pilots to divert the plane to Amarillo.

“Out of an abundance of caution, the pilots in command of the flight diverted to Amarillo, where law enforcement officials met the aircraft and detained six individuals. Additionally, six other members of the group chose to deplane and remain in Amarillo,” Southwest spokesman Brian Parrish said in an email.

The flight landed in Amarillo around 10:45 p.m., and resumed to Chicago at 1:30 a.m., arriving at Midway at 3:22 a.m., about three hours behind schedule.

The men who were detained were being processed at the Randall County Jail and charged with interference with a flight crew, police said. They have been identified as Saiman Hermez, 19; Jonathan Khalid Petras, 20; Ghazwan Asaad Shaba, 21; Essa Solaqa, 20; Khalid Yohana, 19; and Wisam Imad Shaker, 23. They are all from the San Diego area.

Good morning HAL,

I use that term all the time. I didn't mean it literally.

My Mom used to say, "You weren't raised outdoors, stop acting like you were."

Another favorite of hers was, "I didn't raise my kids to act like that!" grin
